PREGABALIN

Indications & Dose:

Peripheral and central neuropathic pain



Adult: Initially 150 mg daily in 2–3 divided doses, then increased if necessary to 300 mg daily in 2–3 divided doses, dose to be increased after 3–7 days, then increased if necessary up to 600 mg daily in 2–3 divided doses, dose to be increased after 7 days


Adjunctive therapy for focal seizures with or without secondary generalisation



Adult: Initially 25 mg twice daily, then increased in steps of 50 mg daily, dose to be increased at 7 day intervals, increased to 300 mg daily in 2–3 divided doses for 7 days, then increased if necessary up to 600 mg daily in 2–3 divided doses


Generalised anxiety disorder



Adult: Initially 150 mg daily in 2–3 divided doses, then increased in steps of 150 mg daily if required, dose to be increased at 7 day intervals, increased if necessary up to 600 mg daily in 2–3 divided doses  

Contraindications:

Hypersensitivity to the active substance or to any of the excipients

Side Effects:

The most commonly reported adverse reactions were dizziness and somnolence. The common (> 1/100, < 1/10) adverse effects associated with pregabalin: appetite increased, euphoric mood, confusion, libido decreased, irritability, ataxia, disturbance in attention, coordination abnormal, memory impairment, tremor, dysarthria, paraesthesia, vision blurred, diplopia, vertigo, dry mouth, constipation, vomiting, flatulence, erectile dysfunction, weight increased, fatigue, edema peripheral, feeling drunk, edema, gait abnormal

Since pregabalin is predominantly excreted unchange in the urine, undergoes negligible metabolism in humans «2% of a dose recovered in urine as metabolites), does not inhibit drug metabolism invitro, and is not bound to plasma proteins, it is unlikely to produce, or be subject to, pharmacokinetic interactions.

There are no adequate data on the use of pregabalin in pregnant women. Breast-feeding is not recommended during treatment with pregabalin. 

Special Precautions:

SPECIAL WARNINGS AND SPECIAL PRECAUTIONS FOR USE: 


Patients with rare hereditary problems of galactose intolerance, the Lapp lactase deficiency or glucose-galactose malabsorption should not take this medicine. Some diabetic patient who gain weight on pregabalin treatment may need to adjust hypoglycemic medications. Pregabalin treatment has been associated with dizziness and somnolence, which could increase the occurrence of accidental injury (fall) in the elderly population. Therefore, patients should be advised to exercise caution until they are familiar with the potential effects of the medication
