ESOMEPRAZOLE

Indications & Dose:

BY MOUTH:

• NSAID associated gastric ulcer:

20 mg once daily, for 4-8 weeks

     • Zollinger Ellison Syndrome:

40 mg twice daily, adjusted according to response; usual range 80-160mg (Above 80mg in two divided doses.

• Healing of erosive esophagitis:

20-40 mg one daily for 4-8 wk., for patients who do not heal after 4-8 wk, and additional 4-8 wk of treatment maybe considered

• Maintenance of healing of erosive esophagitis:

20 mg qd

GERD:

20-40mg one daily for 4 wk: if symptoms do not resolve completely after4 wk, an additional 4wk of treatment my he considered maintenance 20mg one daily.

 Eradication of H. pylori:

40 mg qd in combination with amoxicillin 1 g bid and clarithromycin 500 mg bid for 10 days (triple therapy)

Dose adjustment in severe liver impairment:

a dose of 20 mg qd should not be exceeded

Children:
Children: Over 12 years in Gastro-esophageal reflux disease 40mg once daily for 4 weeks, continue for further 4 weeks if not fully healed.,maintainace 20mg daily.

Children: -1-12 years , body weight 10-20kg, 10mg once daily for 8 weeks, body weight over 20 kg 10-20 mg once daily for 8 weeks

BY INJECTABLE

By intravenous injection: over atleast 3 minutes or by intravenous infusion adult over 18 years ,

Gastroesophageal diseae

40mg once daily. Severe peptic ulcer bleeding :(following endoscopic treatment adult over 18 years initial infusion 80 mgover 30minutes/hour for 72 hours.
other conditions :20mg continue till oral administration possible

Contraindications:

CNS: Asthenia. dizziness, headache
GI: Abdominal pain, constipation, diarrhea, flatulence, nausea, dry mouth
MS: Back pain
RES: Cough
SKIN: Rash

Drugs

• Ketoconazole, iron salts, digoxin: Reduced gastric acidity may result in decreased absorption of these and other drugs where gastric pH is an important determinant of bioavailability.
