Medroxyprogestrone Acetate

Indications & Dose:

Contraception, menstrual problems, endometriosis. Palliative treatment of some hormone dependent neoplasms such as endometrial carcinoma, breast cancer hypernephroma, prostate carcinoma, treatment of prostate hypertrophy.

• BY INJECTION

Inj: 150mg by deep i.m. inj during 1st 5 days of cycle, repeat at 12 weeks intervals.

Post partum, 150mg by deep i.m. inj within first 5 days if not breastfeeding or after 6 weeks if breastfeeding repeat at 12 weks intervals.

As long as the inj. is given no later than 5 days after the 12 weeks interval no additional contraception required. If later, pregnancy must be excluded before injection, is repeated and barrier contraception then used for 14 days.

• BY MOUTH

Tab: Breast carcinoma: 0.4-1.5 mg daily

Endomerial and renal carcinoma:

0.200-600 mg daily

Prostatic carcinoma:

100-600mg daily

Contraindications:

Pregnancy, undiagnosed vaginal bleeding, hepatic impairment or active liver disease, severe arterial disease, breast or genital tract carcinoma, porphyria. 

Side Effects:

Acne, urticaria, fluid retention, weight changes, gastro-intestinal distur-bances, changes in libido, breast discomfort, premenstrual symptoms, irregular mens-trual cycles; also depression, insomnia, somnolence, alopecia, hirsutism, anaphylactoid-like reaction; rarely jaundice. Transient infertillity on continous treat-ment, irregular, prolonged or heavy vaginal bleeding during first 2 or 3 cycles. Back pain, weight gain, fluid retention. 

Cautions:

 Diabetes, hypertension, cardiac or renal disease. 

Precautions:

Undiagnosed abnormal vaginal bleeding, severe depression, diabetes. While using as contraceptive wait six weeks post-partum if lactating.

Carbamezapine, phenylbutazone, barbiturates, phenytion, primidone, rifampicin, griseofulvin, tetracycline
