Solifenacin Succinate

Indications & Dose:

Treatment of overactive bladder with symptoms of urinary frequency, urgency, or urge incontinence

• BY MOUTH

Adult:  5mg once daily  once daily increase to 10mg once daily.
Children:  Not recommended 

Contraindications:

Hypersensitivity to solifenacin or any component of the formulation:urinary retention: gastric retention; uncontrolled narrow-angle glaucoma

Side Effects:

Gastrointestinal: Xeroatomie , constipation
Cardiovascular: Edema, hypertension

Central nervous system: Dizziness, fatigue , depression .
Gastrointestinal: Nausea . dyspepsia, upper abdominal pain, vomiting.
Genitourinary: Urinary tract infection, urinary retention.
Ocular: Blurred vision , dry eyes
Respiratory: Cough , pharyngitis.
Miscellaneous: Influenza

Cytochrome P450 Effect: Substrate of CYP3A4 (major) Increased Effect/ toxicity: CYP3A4 inhibitors may increase the levels/ effects 01 solifenacin: example inhibitors include antifungals, ciprofloxacin, clarithromycin. diclotenaz, doxycycline, erythromycin, ketoconazole, imatinib, isoniazid, nefazodore. nicardipine. propofol, protease inhibitors, quinidine and verapamil; solifenacin dose should not exceed 5 mg/day.

Decreased Effect:

CYP3A4 inducers may decrease the levels/effects of solifertacas example inducers include aminoglutethimide, carbamazepine, nafcillin. nevirapine, phenobarbital, and phenytoin.

Ethanol/Nutrition/Herb Interactions

Food: Grapefruit juice may increase the serum level effects of solifenacin.

Herb/Nutraceutical: St Johns wort (Hypericum) may decrease the levels/effects ci solifenacin. Stability Store at room temperature between 15°C to 30°C (59°F to 86°F). 

Warnings:

Use with caution in patients with bladder outflow obstruction, gastrointestinal obstructive disorders, and decreased gastrointestinal motility. Use with caution in patients with controlled (treated) narrowangle glaucoma.
