DIDANOSINE

Indications & Dose:

INDICATIONS AND DOSE:

REV infection in combination with other antiretroviral drugs. 

 • BY MOUTH

• ADULT under 60kg 250mg daily in 1—2 divided doses, 60kg and over 400mg daily in 1—2 divided doses;

CHILD over 3 months (under 6 years Videx tablets only), 240 mg/m2 daily (180 mg/m2 dairy in combination with zidovudine) in 1—2 divided doses.

Side Effects:

see notes above; also pancreatitis (see also under cautions), liver failure, anaphylactic reactions, peripheral neuropathy—suspend (reduced dose may be tolerated when symptoms resolve), diabetes mellitus, hypoglycaemia, acute renal failure, rhabdomyolysis, dry eyes, retinal and optic nerve changes (especially in children), dry mouth, parotid gland enlargement, sialoadenitis, alopecia, hyperuricemia (suspend if raised significantly) 

Cautions:

History of pancreatitis (preferably avoid, otherwise extreme caution, see also below); peripheral neuropathy or hyperuricaernia (see under Side-effects); dilated retinal examinations recommended (especially in children) every 6 months, or if visual changes occur; interactions: (didanosine) Pancreatitis Suspend treatment if serum lipase raised (eve if asymptomatic) or if symptoms of pancreatitis develop; discontinue if pancreatitis confirmed. Whenever possible avoid concomitant treatment with other drugs known to C cause pancreatic toxicity (e.g. intravenous pentamidine isetionate); monitor closely if concomitant therapy unavoidable Since significant elevations of triglycerides cause pancreatitis monitor closely if elevated
