PEFLOXACIN

Indications & Dose:

Respiratory tract, urinary tract, skin and soft tissue, surgical, bone and joint, gynaecological infections. Sexually transmitted, gastrointestinal and severe systemic infections. Gonorrhoea, Septicaemia, osteomyelitis and meningitis. 


DOSE



Adults


Tab: With normal liver function: 400mg twice daily. An initial loading dose of 800mg may be given in order to produce effective blood concen-trations more rapidly.



Gonorrhoea: 800mg single dose. Inj: Admin. by slow i.v. inj. at a dose of 400mg diluted in 100-250ml of 5 per cent isotonic glucose solution (over a period of 1 hour), twice daily.


Children: Not recommended.

Contraindications:

 Pregnancy, breastfeeding,  G6PD deficiencies. Hypersensitivity, children below 15 years.


 

Side Effects:

Nausea, epigastric distress, arthralgia, skin rash, photosensitivity reaction, insomnia, headache, myalgia and thrombocytopenia.

Hepatic dysfunction, avoid exposure to sunlight during treatment.


Old age: May be used. 

Interaction:

Drugs


Antineoplastic drugs: Decrease serum levels of pefloxacin.


Caffeine: Enhanced efficacy of caffeine.


Oral anticoagulants: Enhanced efficacy of anticoagulants.


Cyclosporine: Nephrotoxicity increased by pefloxacin.


Theophylline: Increased plasma levels of theophylline resulting in toxicity.


NSAIDs: CNS excitation may occur.


Rifampicin: Decreases serum concentration of pefloxacin.


Chloramphenicol: Antagonizes effects of pefloxacin
