OFLOXACIN

Indications & Dose:

Lower respiratory tract infections, uncomplicated urethral and cervical gonorrhea; non-gonococcal urethritis, cervicitis; skin and skin structure infections, UTIs, prostatitis; superficial ocular infections involving the conjunctiva or cornea (ophthalmic preparation)

• BY MOUTH

Dose depend upon type and severity of infection, Usually 200-400 mg once daily or morning or 400- 800 mg daily as in two divided doses at equal intervals for 5-10 days Urinary tract infections: 200-400mg daily preferably in the morning increased if necessary in upper urinary tract infections to 400mg twice daily.

Lower respiratory tract infections : 400mg daily for 7- 10 days preferbly in the morning increased if necessary to 400mg twice. Uncomplicated gonorrhoea: 400mg as a single dose. Non gonococcal urethritis and cervicitis : 400mg daily in single or divided doses, Prostatis: 300mg twice a day upto 6 weeks.

Enteric fever 400mg daily in single or divided doses. By intravenous infusion

• BY INTRAVENOUS INFUSION

Usually 200-800 mg daily by slow over 30 minutes (over at least 30 minutes) complicated     urinary tract infection 200mg twice daily. LRTI / Septiceamia 200mg twice daily.

Severe or complicated infections dose may be increased to 400mg twice daily.

Skin & Soft tissue infections 400 mg daily

Children: Not recommended.

Drugs

Aluminum: Reduced absorption of ofloxacin; do not take within 4 hr of dose

Antacids: Reduced absorption of ofloxacin; do not take within 4 hr of dose.

Calcium: Reduced absorption of ofloxacin; do not take within 4 hr of dose

Iron: Reduced absorption of ofloxacin; do not take within 4 hr of dose

Magnesium: Reduced absorption of ofloxacln; do not take within 4 hr of dose

Procainamide: Ofloxacin competitively inhibits renal tubular excretion of procainamide

Sodium bicarbonate: Reduced absorption of ofloxacin; do not take within 4 hr of dose

Sucralfate: Reduced absorption of ofloxacin; do not take within 4 hr of

Warfarin: Inhibits metabolism of warfarin; increases hypoprothrombinemic response to warfarin

Zinc: Reduced absorption of ofloxacin; do not take within 4 hr of dose

Excreted into breast milks in quantities approximating maternal plasma concentrations; due to the potential for arthropathy  and osteochondrosis, use extreme caution in nursing mothers

CNS: Anxiety, depression, dizziness. fatigue, headache, insomnia, seizures, somnolence

EENT: Dizziness, visual disturbances

GI: Abdominal pain, anorexia, diarrhea, dry mouth; flatulence; heartburn, increased AST, ALT; nausea, pseudomcmbranous colitis, vomiting

SKIN: Photosensitivity, pruritus, rash
