Indications & Dose:
H. influenzae, E. coli, P. mirabilis and N. gonorrhoeae. Gram-positive organisms Streptococci (including Strepto-coccus-faecalis), D. pneumoniae and nonpenici-llinase-producing staphylococci. Therapy may be instituted prior to obtaining results from bacteriological and susceptibility studies to determine the causative organisms and their susceptibility to amoxicillin. Indicated surgical procedures should be performed.
DOSE
Adult
Orally: 500mg-1gm every 6 hourly or more frequently according to the severity of infection.1/ m inj: 500mg-1gm every 4-6 hourly,i/v infusion: in severe infection in adult up to 12gm may be administered.
Children Orally: Neonates: 0.6ml of drops every 4 hourly,1 month-2 years 1/4 of adult dose, 2-10 years 1/2 of the adult dose., i/m, i/v, i/v infusion:
Neonates 75mg every 8 hourly,1 month-2 years 1/2 of the adult dose, 2-10 years, 1/2 of the adult dose.
Contraindications:
A history of allergic reaction to any of the penicillins
Side Effects:
Nausea, vomiting and diarrhoea. Erythematous maculopapular rashes and urticaria have been reported. NOTE: Urticaria, other skin rashes and serum sickness-like reactions may be controlled with antihistamines and, if necessary, systemic corticosteroids. Whenever such reactions occur, drug should be discontinued. A moderate rise in serum glutamic oxaloacetic transaminase (SGOT) has been noted, but the significance of this finding is unknown. Anemia, thrombocytopenia, thrombocytopenic purpura, eosino-philia, leukopenia and agranulocytosis have been reported during therapy with penicillins. These reactions are usually reversible on discontinuation of therapy and are believed to be hypersensitivity phenomena. Reversible hyperactivity, agitation, anxiety, insomnia, confusion, behavioral changes, and/or dizziness have been reported rarely.
Cautions:
As with any potent drug, periodic assessment of renal, hepatic and hematopoietic function should be made during prolonged therapy.The possibility of superinfections with mycotic or bacterial pathogens should be kept in mind during therapy. If superinfections occur (usually involving Enterobacter, Pseudomonas or Candida), the drug should be discontinued and/or appropriate therapy instituted.
