CLORAZEPATE DIPOTASSIUM

Indications & Dose:

Anxiety disorders, acute alcohol withdrawal, partial seizures (adjunctive therapy)



Adult 7.5-22.5 mg daily in 2-3 divided doses or a single dose of 15 mg at bedtime


Elderly (or debilitated) half adult dose


Children 9-12 years initially: 3.75-7.5mg/dose twice daily, increase 3.75mg at weekly intervals. Not exceed to 60mg/day. 

Contraindications:

Narrow-angle glaucoma, psychosis

Interaction:

Cimetidine: Increased plasma levels of clorazepate and/or active metabolites


Disulfiram: Increased serum clorazepate concentrations


Ethanol: Enhanced adverse psychomotor side effects of benzodiazepines


Rifampin: Reduced serum clorazepate concentrations

 Excreted into breast milk; drug and metabolites may accumulate to toxic levels in nursing infant

CNS: Anterograde amnesia, anxiety, ataxia, confusion, depression, dizziness, drowsiness, fatigue, hallucinations, headache, hysteria, insomnia, psychosis, slurred speech, stimulation, tremor


CV: Bradycardia, cardiovascular collages, hypertension, hypotension, orthostatic, tachycardia


EENT: Auditory disturbances, blurred vision, mydriasis, nystagmus


GI: Anorexia, constipation, diarrhea, dry mouth, nausea, vomiting


GU: Changes in libido, incontinence, menstrual irregularities, urinary retention


SKIN: Dermatitis, hair loss, hirsutism, itching, rash, urticaria
